The Bluestone sculpin (Cottus sp.1) is an undescribed freshwater fish endemic to the New River system in Virginia and West Virginia. The taxonomic status of the Bluestone sculpin is unresolved, and its range often overlaps with congeners, with which it may hybridize. The species is most often found in small, cool streams with gravel and rubble dominated substrates. Juveniles and adults are found most often in runs and riffles of boulder and rubble (Jenkins and Burkhead 1994). Many of the streams presumed to be within the species’ range run along highways and may be vulnerable to non-point source pollution. Biological indicator taxa have long been used for integrative assessments of water quality, particularly benthic invertebrate groups such as arthropods. While standardized protocols have been developed to calculate 'biological index' scores based on the abundances of these taxa, such systems are challenging to implement at large scales due to the sampling effort required, taxonomic expertise needed, and the need for repeated sampling to reliably discriminate sites. The Grand Calumet River flows through northwestern Indiana and was designated an EPA Area of Concern (AOC) in 1987. There are multiple legacy pollutants in the river including but not limited to heavy metals and PCBs. To investigate potential causes and patterns of eutrophication in the river, water samples were collected for 16S rRNA microbial and eukaryotic 18S rRNA amplicon sequencing during April - September 2021. This data release describes the resulting sequencing data.

The data being released were part of a project funded by the United States Environmental Protection Agency (USEPA). This study sought to identify the planktonic communities (cyanobacteria, eukaryotic algae) potentially contributing to eutrophication within the Grand Calumet River Area of Concern (AOC) in northern Indiana along the southern shore of Lake Michigan. In 2021, triplicate water samples were collected from five locations during three events, 4/19/21 and 4/20/21, 7/7/21, and 9/15/21. Data were collected to determine the abundance of nitrogen-fixing microorganisms in Cladophora algae growing on rocks, breakwall structures, or submerged dreissenid mussel beds around southern Lake Michigan. Cladophora samples (N=33) were collected between June and September 2015 from three urban areas: (a) Jeorse Park, East Chicago, Indiana, (b) Calumet Beach, Chicago, Illinois, and (c) North Beach, Racine, Wisconsin, and a National Park site, Portage Lakefront, Indiana Dunes National Park, Indiana. The FWS considers the Appalachian grizzled skipper (AGS, Pyrgus wyandot) a Species of Concern and is requesting key information on this species for a status review. According to the RFP, FWS has given AGS full species status, however, AGS’s taxonomic status is considered controversial. Some lepidopterists recognize P. wyandot as a full species whereas others consider it a subspecies, P. centaureae wyandot (pers. comm. B. Leppo, PA Natural Heritage Program; in Chazal et al., 2004). Regardless, FWS can still list AGS as threatened or endangered at the species or subspecies level. The Deep SEARCH Project (Deep sea exploration to advance research on cold corals/canyon/cold seep habitats) included a cruise in April 2019 aboard the NOAA ship Ron Brown to various sites along the Mid-Atlantic North American coast. An ROV was used to collect samples of bottom water at these sites, which were then subjected to eDNA extraction and analyses. This data release describes what samples were collected for eDNA analyses, and how eDNA was extracted, sequenced, and bioinformatically analyzed.

Environmental DNA water samples were collected at multiple sites in Great Smoky Mountains National Park in 2016 and 2017 and analyzed with a droplet digital PCR assay to screen for the presence of smoky mountain madtoms. Smoky Mountain madtoms were detected within a section of Abrams Creek, known as the only stream in Great Smoky Mountains National Park that supports this species.
